The thiazide diuretics enter the lumen of the nephron by two routes (click to see the routes).
Within the distal convoluted tubule the thiazides bind to the Cl- site and thus inhibit the luminal Na+/Cl- co-transporter. The reabsorption of Na+, Cl- and water is therefore inhibited. The volume of nascent urine carried onwards to the collecting ducts is greater than normal and is rich in Na+ and Cl- (click to see this process).
